Bacterial Pathogens in Seafood ArchiMer

A number of bacterial illnesses may arise from the consumption of seafood that has either been contaminated at source or which becomes contaminated during the processing and retail chain. Such illnesses may arise from infection with the bacteria themselves or by the ingestion of toxins formed in the foodstuff prior to consumption. This division is actually too simplistic: for example, the toxins of Clostridium botulinum and Staphylococcus aureus are preformed in food during bacterial growth, the toxin of Clostridium perfringens is usually only formed when the bacteria sporulate in the intestinal tract while the toxin of Vibrio cholerae O1 (and O139) is produced when the bacteria multiply in the intestinal tract.

In the marine environment, biofilm formation is an important lifestyle for microorganisms. A biofilm is comprised of cells embedded in an extracellular matrix that holds them close together and keeps the biofilm attached to the colonized surface. This predominant lifestyle and its main regulation pathway, namely quorum-sensing (QS), have been shown to induce specific bioactive metabolites. Detection of cytotoxin-hemolysin mRNA in nonculturable populations of environmental and clinical Vibrio vulnificus strains in artificial seawater ArchiMer

The objective of this study was to develop a molecular detection method that better estimates the potential risk associated with the presence of Vibrio vulnificus. For that purpose, we applied seminested reverse transcription-PCR (RT-PCR) to viable but nonculturable (VBNC) populations of V. vulnificus and targeted the cytotoxin-hemolysin virulence gene vvhA. Discrimination between human, pig and ruminant fecal contaminations in a river catchment by real-time PCR using host-specific markers ArchiMer

The microbiological quality of coastal waters and shellfish harvesting areas in Brittany (France) can be affected by faecal pollutions from human activities and animal breeding (especially pigs and cattle). To discriminate among faecal pollution of human and animal origin, a library-independent microbial source tracking method was selected: Bacteroidales host-specific 16S rRNA gene markers by real-time Polymerase Chain Reaction (PCR). A human-specific Bacteroidales marker (Hum-1-Bac) was designed. Tested on faecal samples, the Hum-1-Bac marker showed 95 % sensitivity and 95 % specificity (n= 80). Ecology of pathogenic and non-pathogenic Vibrio parahaemolyticus on the French Atlantic coast. Effects of temperature, salinity, turbidity and chlorophyll a ArchiMer

Vibrio parahaemolyticus is one of the principal bacterial causes for seafood-borne gastroenteritis in the world. In the present study, three sites located on the French Atlantic coast were monitored monthly for environmental parameters over 1 year. The presence of total and pathogenic V. parahaemolyticus in sediment, water and mussel samples was detected following enrichment by culture and real-time PCR (toxR gene, tdh, trh1 and trh2 virulence genes). Since the spring of 2014, the mussel farming of Charente-Maritime and Vendée (French Atlantic coast) has experienced an unprecedented crisis. Mass mortality events of blue mussel (M. edulis) have affected livestock areas, decimating up to 100% of production in some farming areas. Investigation for infectious agents in moribund mussels revealed presence of bacteria capable of inducing mortalities. Vibrio spp. are ubiquitous marine bacteria that are ecologically and metabolically diverse members of planktonic and animal associated microbial communities. They encompass the ancient and well-studied human pathogen, Vibrio cholerae, and two other human pathogens, V. vulnificus and V. parahaemolyticus, as well as some less thoroughly characterized animal pathogens. Infectious diseases linked to the consumption of raw shellfish like oysters, mussels, cockles and clams, have long been identified. Bacterial diseases such as cholera and typhoid fever were the first to be suspected of being linked to consumption of contaminated shellfish (Butt et al., 2004). During the past century, various strategies have been established in shellfish growing areas throughout the world to assure the sanitary quality of shellfish. More recently, despite sanitary surveys, Vibrio parahaemolyticus, Vibrio vulnificus and enteric viruses - especially Hepatitis A virus (HAV) and norovirus (NoV) – were found to be associated in outbreaks of human illness.

Real-time reverse transcription-PCR for transcriptional expression analysis of virulence and housekeeping genes in viable but nonculturable Vibrio parahaemolyticus after recovery of culturability ArchiMer

A real-time reverse transcription-PCR method was developed to determine whether the recovery of culturability of viable but nonculturable (VBNC) Vibrio parahaemolyticus induced the expression of virulence genes coding for the thermostable direct hemolysin and for type III secretion system 2 (TTSS2). The culturability of clinical strain Vp5 of V. parahaemolyticus in artificial seawater at 4°C was monitored, and the VBNC state was obtained. Recovery in culture of viable but nonculturable Vibrio parahaemolyticus: regrowth or resuscitation? ArchiMer

The objective of this study was to explore the recovery of culturability of viable but nonculturable ( VBNC) Vibrio parahaemolyticus after temperature upshift and to determine whether regrowth or resuscitation occurred. A clinical strain of V. parahaemolyticus Vp5 was rendered VBNC by exposure to artificial seawater ( ASW) at 4 degrees C. Aliquots of the ASW suspension of cells ( 0.1, 1 and 10 ml) were subjected to increased temperatures of 20 degrees C and 37 degrees C.
